anti-Cyclin-Dependent Kinase 4 mouse monoclonal, DCS-156, lyophilized, purified
Western blot analysis of human HeLa cell lysate with anti-Cyclin-Dependent Kinase 4 antibody. Western blot analysis was performed on either 5 µg or 10 µg of HeLa lysate. Cells were lysed in PBS by homogenization. The PVDF membrane was blocked with 5% dry milk in PBST for 1 h at RT. The primary antibody anti-Cyclin-Dependent Kinase 4 mouse monoclonal, DCS-156 (Cat. No. 690096) was diluted in blocking buffer (antibody concentration 0.5 µg/ml) and incubated for 1 h at RT. The secondary antibody goat anti-mouse IgG polyclonal, HRP conjugate was also diluted in blocking buffer (antibody concentration 0.2 µg/ml) and incubated for 1 h at RT. The bands were visualized by chemiluminescent detection using PierceTM ECL Western Blotting Substrate.

Background
DCS-156 reacts specifically with cdk4 protein present predominantly in G1 phase of cell cycle; it does not cross-react with cyclins. In immunohistochemical application on frozen sections of head and neck carcinoma (oropharynx and hypopharynx carcinoma) and cervix DCS-156 shows distinct nuclear staining, especially in tumor areas of enhanced proliferation.
Epitope recognized: aa 270-290.

PROGEN offers different formats for mouse monoclonal antibodies. Different antibodies may not be offered in each format, the following are all possible formats:
- Supernatant and supernatant concentrate: This format contains hybridoma cell culture supernatant. The antibody is not purified and the antibody concentration is not determined. The antibody concentration may vary from lot to lot. Therefore we recommend to titrate the optimal concentration for the application used for each new lot.
- Lyophilized, purified: This format contains purified antibody in lyophilized form. The reconstitution of this antibody is described in the datasheet. The buffer composition after reconstitution is also mentioned on the datasheet.
- Liquid, purified: This format contains purified antibody in liquid format. The concentration is mentioned on the datasheet.
- Prediluted, purified: This format contains purified antibody in liquid format. Most antibodies in this format are diluted to be ready-to-use for IHC with standard tissue. But some antibodies of this format need further dilution for IHC. This is mentioned on the datasheet.
Lyophilized antibodies can be stored at 2-8°C until expiration.
