Antibodies for the detection of AAV

PROGEN antibodies are useful for the characterization of different stages of an AAV infection and effective in the analyses of the AAV assembly process. There are two main categories of AAV antibodies:

  1. AAV capsid protein antibodies that specifically react with denatured AAV capsid proteins and thereby recognize the linear epitopes of the viral capsids.
  2. AAV particle antibodies that specifically react with intact AAV particles, i.e. empty and full capsids and thereby recognize a conformational epitope of the assembled capsids of certain serotypes.

AAV Capsid Protein Antibodies

The AAV capsid protein antibodies recognize linear epitopes of the viral capsid proteins that consists of neighboring amino acids. These epitopes have high sequence similarity and therefore the antibodies detect the viral capsid proteins VP1, VP2 and VP3 across the serotypes.

On the left hand side you can see the primary sequences of the three capsid proteins and the binding sites along the sequence. Since all proteins are encoded by the same gen they only differ in alternate splicing. Therefore antibody B1 is able to bind all capsid proteins whereas the binding site of antibody A1 is only located on VP1. For the same reason antibody A69 is only suitable for VP1 and VP2.

All of the antibodies are useful to study the expression levels of VP1, VP2 and VP3 in western blot analyses, in order to determine the protein localizations or protein-protein interactions by affinity chromatography, immunoprecipation or immunofluorescence.

AAV Particle Antibodies

The AAV particle antibodies recognize conformational epitopes of the viral capsid proteins that are only present on fully assembled capsids. These epitopes consist of distant amino acids either of the same protein or of different proteins that are in close proximity, and they might vary across the different serotypes. However, for some of the antibodies some cross-reactivity is known (for more information, please see the specific AAV particle antibodies by AAV serotype below). The AAV particle antibodies are useful for the following processes:

  • to study fully assembled and intact capsids by dot blot analyses
  • to quantify the intact viral capsids (empty and full) by ELISA
  • to detect neutralizing antibodies in patients serum
  • to determine protein-protein interaction or to purify virus preparations by affinity chromatography or immunoprecipitations
  • to analyze virus particle localizations by immunofluorescence.
  • Some of the following antibodies are also available in sample sizes of 10µg (lyophilized). 

AAV Replicase Antibodies

PROGEN also offers antibodies reacting with the four regulatory proteins Rep78, Rep68, Rep52 and Rep40. These proteins are encoded by the rep-gene, playing an important role in AAV genome replication and packing. During AAV Infection mentioned proteins are always expressed in different concentrations in human cells. Check out the table below which antibody suits your demand.

PerkinElmer has integrated PROGEN’s well-established AAV2 particle antibody A20R into their no-wash AlphaLISA® assay technology. The AlphaLISA® AAV2 Capsid Detection Kit is designed to quantify AAV2 capsids in buffer, cell culture media, cell lysates and cell supernatants.

PROGEN’s A20R antibody exclusively detects fully assembled, intact AAV2 capsids, providing an efficient and reliable quantification of the overall AAV2 capsid titer.
